Her career began with Indian television, where she starred in Mujhse Kuchh Kehti…Yeh Khamoshiyaan (2012–2013) and gained widespread recognition with Kumkum Bhagya (2014–2016). Her performance earned her the Indian Television Award for Best Supporting Actress.

Thakur transitioned to films with the Indo-American drama Love Sonia (2018), which tackled human trafficking and showcased her ability to handle intense roles. She quickly rose to prominence in Bollywood with Super 30 (2019), opposite Hrithik Roshan, and Batla House (2019), both of which were commercial successes.

After a few mixed box-office performances, she reinvented her career in Telugu cinema. Her role in Sita Ramam (2022), opposite Dulquer Salmaan, was a turning point, earning her critical acclaim and multiple awards, including SIIMA Best Actress (Critics). She followed this success with Hi Nanna (2023), further solidifying her position in South Indian cinema.
