Born on December 13, 1990, in Chennai, Tamil Nadu, Regina Cassandra hails from a multicultural background. Her father is North Indian, while her mother is Kannada, and her grandmother is a Goan Anglo-Indian. She developed an interest in performing arts at a young age and started anchoring for a kids' TV show when she was just nine years old. Regina pursued a Bachelor of Science in Psychology from Women’s Christian College, Chennai, before fully committing to her acting career.

Regina made her acting debut in the Tamil film Kanda Naal Mudhal (2005), where she played the role of Latha, a 21-year-old pregnant girl. Her performance was well-received, and she later ventured into Telugu cinema, making her debut with Siva Manasulo Sruthi (2012), which won her the SIIMA Award for Best Female Debut – Telugu.
